Overview of MSWS Control of 212-LC July 15, 2006

2 Software Overview Functionality Derived from ProStar/Dynamax system driver used for 210s Only supports Binary system at this point. RS232 connection to Pump A only, requires port in COM1-4 range Method flow and composition ramps (up to 100 steps). Optional ramp on method activation and completion Manual mode GLP features (serial numbers, solvent info, stroke counters) Distribution 212-LC driver available for MSWS 6.6 SP1 and above “212-LC Control Software” CD to add driver to MSWS 6.6 SP1 ships with pumps – only needed with MSWS 6.6 SP1. Driver included in “LC Drivers” in MSWS 6.7 and above (installed when selecting (1200 LC or 500-MS during installation)

3 212-LC Software Configuration (1) In System Control, select Instruments->Setup Comm Ports… For the serial port to which the 212- LC System is connected, pull down the Combo Box and select 212-LC

4 212-LC Software Configuration (2) In the configuration screen, the 212-LC module icon appears in the ‘Available Modules’ area. Drag it to the instrument window as shown. Switch to the Instrument screen and open the 212-LC window.

5 212-LC Software Configuration (3) In the 212-LC Control and Status window, push the ‘System Setup’ button. The System Setup dialog shows the current configuration and allows you to set solvents and change Pmin & Pmax Clicking on one of the Min or Max Pressure cells enables the ‘Set Pressure Limits’ button. Clicking on one of the ‘Solvent’ cells enables the ‘Select Solvent’ button. Check boxes control whether or not composition and pressure traces are displayed.

6 212-LC Software Configuration (4) In ‘System Setup’, click on one of the cells showing Pressure Limits Press the ‘Set Pressure Limits’ button. Make a meaningful change to Pmax and press OK The selected values will be set for both pumps. In ‘System Setup’, click on the name of Solvent B Press the ‘Select Solvent’ button. Select a new solvent from the list and press ‘OK’. Press ‘Save’ to exit the System Setup dialog. Depending on changes made, a warning will come up saying that you need to change the Active Method or activate a new method. Opt to edit the active method.

7 212-LC Method Editor When the method editor opens up, select the 212-LC section As you do so, the editor may detect that the configuration does not match that of the instrument last connected at that address and offer to update the method. Press ‘Yes’.

9 Method Editor: Time Program Spreadsheet Click and type in cells to edit Enter time as mm:sec or fractional minutes (0:30 or 0.5) Enter %B, %A is computed Enter flow in uL/min “Blank” value means same as previous Option to show or Hide %A

10 Method Editor: Time Program Edit Buttons Basic Editing Add, Insert, Delete – all apply to the current selection in the spreadsheet Advanced Editing Scale Run Time: Recompute all the steps to scale the whole program to a different length. Add / Subtract Run Time from specific location in program.

11 Method Editor: Set Configuration Configuration holds Pump Info Solvent info The configuration is used to Know the pump info when editing off-line Verify at download time that the method was meant to be used with the current solvents and pump options

12 Method Editor: Ramps Ramp on Activation Ramp Time – Upon method activation, ramp from the current flow/composition to the time zero conditions of the method. Hold Time – equilibration time to follow the ramp. Ramp on Completion Ramp Time – At the end of the method, ramp from the final flow/composition to the time zero conditions of the method. Hold Time – equilibration time to follow the ramp.

13 System Control: Activation In the 212-LC window, press the ‘Activate’ button. This activates the method and starts the pumps. If a ramp on activation is specified, the window title will read ‘Equilibrating’, then ‘Stabilizing’ while the ramp and hold time are satisfied. During the ramp, the Runtime field counts down from ramp time + hold time. Once Activation is complete, the window title reads ‘Ready’ and the ‘Run’ button is enabled.

14 System Control: Running the 212-LC If you are set up with an AutoSampler, use ‘Inject Single Sample’ to initiate a run. Otherwise, just press the ‘Run’ button in the 212-LC window. As the program executes, the Runtime is updated in min:sec, and the time program and pressure traces are shown if specified in ‘System Setup’.. At the end of the run, the display is ‘frozen’ until the start of the next run. Press ‘Resume Plot’ to un- freeze the display.

15 System Control: ‘Run Status’ controls The ‘Hold’ button is active during the run. It suspends the time program and extends the end time accordingly. Press ‘Continue’ to resume the program. See trace for held/resumed program below. The ‘Method’ button calls the method editor with the module’s method section selected. When the editor is closed, System Control will offer to reactivate the module method if it has been modified..

16 System Control: ‘Pump Status’ and Keypad The ‘Unlock’ button temporarily disconnects the pumps and returns functionality to the pump’s keypad. Press ‘Lock’ to reconnect the pumps. ‘Hide Keypad’ hides most of the keypad to give more space to the graphics. Press ‘View Keypad’ to restore it. STOP PUMPS immediately stops the pumps.

17 System Control: 212-LC Manual Mode In the 212-LC Window, press ‘Manual Control’ Select the desired composition by editing %B Edit the desired flow and ramp time. If no ramp is needed, set the time to zero. Press ‘Start Ramp’ to execute the command. e system will ramp flow and composition from the original settings to the new settings. When done with the Manual Mode, press ‘End Manual Control’

18 System Control: GLP Information Displayed as a result of pressing the ‘GLP’ button in the 212-LC window. Keeps track of serial numbers, firmware versions, solvent info, and stroke counters. The bottom three counters are associated with consumable items and can be reset when the items are replaced. The GLP information is stored in each data file produced by the instrument including the 212-LC System.

LC Run Log MS and Chromatography data files acquired by an instrument include a full copy of the instrument method. The 212-LC method used can be seen in the Run Log in the Standard Report Application or in the MS Data Review Application.
