RNA N6‐methyladenosine methyltransferase‐like 3 promotes liver cancer progression through YTHDF2‐dependent posttranscriptional silencing of SOCS2 Knockdown of METTL3 significantly suppressed HCC cell proliferation, colony formation, and migration. (A) Stable knockdown of METTL3 in Huh‐7 and HepG2 cells by lentiviral shRNA sequences (shMETTL3#1 and #2). The knockdown effect was verified at both the mRNA and protein levels. (B) Knockdown of METTL3 significantly reduced the proliferation rate of Huh‐7 and HepG2 cells. Mean ± SD. (C) Knockdown of METTL3 impaired colony‐formation ability of HCC cells. (D) Knockdown of METTL3 suppressed HCC cell migration. Bar graphs: Mean ± SD. **P < 0.01, ***P < (t test). Abbreviations: HPRT, hypoxanthine‐guanine phosphoribosyltransferase; shNTC, nontarget control shRNA.
