Comparison of AFP variants in HPLC assay, NMR spectroscopy, and bioactivity assay. (A) AFP isolated by cation exchange chromatography eluted in two fractions (black) denoted AFP1 and AFP2. (B) Reinjection of the fractions resulting in lead chromatograms of AFP1 (blue) and AFP2 (red). Both AFP variants were analyzed by NMR spectroscopy. 1H,15N-SOFAST-HMQC results of experiments exploiting the natural abundance of 15N-atoms were recorded and are presented as overlaid data, with AFP1 marked black and AFP2 marked red.
