Programming and Curriculum Developed with teachers Based on MB curriculum Hands-on, authentic science Grade 5: Maintaining a healthy body Grade 6: Health curriculum Grade 7: Solutions, ecosystems Grade 8: Cells and systems, optics, fluids Grade 9: Reproduction Grade 11 Biology: Body systems Grade 12 Biology: Genetics

Lab Safety Basics If you don’t know what it is, don’t touch it! No food or drinks! Wear safety equipment when instructed! Follow instructions! Ask questions if you don’t understand instructions!!!

Lab Safety Basics You must wash your hands when leaving the lab and after removing your gloves! Report any spills immediately!

Lab Safety Basics You will be wearing gloves for a reason! DO NOT touch other people or any surfaces when wearing gloves!

Your patient Patient was brought in by his wife after he fainted at home. He is complaining of severe chest pain.

You listen to his heart: Sound 1 Sound 2 Tests You take his blood pressure: BP= 168/93

EKG You order an EKG: this can show us if his heart is beating properly. normal patient

Echocardiography Turbulent flow around aortic valve Left ventricular hypertrophy

Diagnosis: Aortic Stenosis Normal aortic valve Stenotic aortic valve

What now? CT Scan Valve replacement surgery

Or could we build him a new valve? 1.Get a donor valve 2.Decellularize donor valve 3.Get stem cells from patient 4.Grow stem cells from patient into the cell type we want

Stem cells Any type of cell! Some types of cells! From bone marrow or possible other sites

Stem cell therapy Grow billions of cells Push to cell type we want

Cell culture Muscle cells (myocyte) Fibroblasts and myofibroblasts Digest Research

1.Using a dropper, suck out the media (liquid cell food), discard in waste beaker 2.Using a clean dropper, cover with PBS and gently wash cells by rocking the dish 3.Remove PBS and discard in waste tube. Using a clean dropper each time, do two more PBS washes and discard. Check cells under scope when done. 4.Using a clean dropper, add 2ml trypsin and rock in an ‘X’ pattern until cells are detached (check on the scope, about 5min) 5.Using a clean dropper, add 2ml DMEM and draw up all of the liquid with the dropper DO NOT PUT INTO THE WASTE CONTAINER!!! 6.Put the liquid into a clean, empty 15ml blue capped tube 7.Wait for instructions to spin in centrifuge Working with live cells

Next steps We hope to partner with your school division! Division consultants are attending the BIOlab in November, please get in touch with yours to express interest. Get in touch with Steve Check out the website
