A, B, the CNE2 cells were treated with CoCl2 (200 μmol/L) or exposed in a hypoxia condition (1% O2) for 24 hours, and the subcellular colocalization and the interaction of TFAP2A with HIF-1α proteins in nasopharyngeal carcinoma cells were analyzed by immunofluorescence assay (A) and immunoprecipitation (B), respectively. C, D, the CNE2 cells were treated with CoCl2 (200 μmol/L) or exposed in a hypoxia condition (1% O2) for 24 hours, and followed treatment with the DOTAP-cholesterol–based TFAP2A siRNA nanoparticles (si-TFAP2A) and/or the HIF-1α siRNA nanoparticles (si-HIF-1α). At 48 hours after treatment, the effects of the TFAP2A and HIF-1α knockdown on VEGF expression (C) and cell viability (D) were determined by Western blot and MTT analyses, respectively. The cells treated with nonspecific scramble siRNA (si-NS) were used as the control groups.
