Effect of SFN on the protein expression of Nrf2, HO-1, and NQO1 in JB6-shMock and JB6-shNrf2 cells. Cells (1 × 105/10-cm dish) were incubated with various concentrations of SFN (0–5.0 μmol/L) for 5 days. Separation of the cellular proteins was achieved using SDS-PAGE and Western blotting, as described in the Materials and Methods. The protein expression of Nrf2 and the Nrf2 target enzymes HO-1 and NQO1 was detected by hybridization with specific antibodies. The relative expression levels were quantified based on the signal intensity of the corresponding bands from 3 independent experiments and were normalized using β-actin. The graphical data are presented as the mean ± SD from 3 independent experiments. * and # represent P < 0.05 and P < 0.01, respectively, which indicate significant differences in the target proteins compared with their levels in JB6-shMock cells without SFN treatment.
