How Does it Work Many different ways First- need to get the gene of interest Ex. Glow gene from jellyfish Second- need to amplify (make more of) gene of interest Polymerase Chain Reaction (PCR)

Steps continued Third- obtain a plasmid (cloning vector) -A plasmid is a small loop of DNA in a bacterium Four- insert gene into plasmid and have bacteria take up plasmid (transformation) Five- grow bacteria and harvest the gene or protein!

What does all the DNA cutting? Restriction enzymes Cut at particular sequences Cuts gene of interest out and the plasmid at same bases Sticky ends are what is typically produced! To seal the two together, you need ligase! Then you have recombinant DNA
