Introduction to the Microscope Care Parts Focusing Wet mounts Calculating magnification
3
Eyepiece Body Tube Revolving Nosepiece Arm Objective Lens Stage Stage Clips Coarse Focus Fine Focus Base Diaphragm Light
4
Always carry with 2 hands- one hand on arm – other on base Only use lens paper for cleaning Do not force knobs
5
1/Use the coarse adj. to raise the body tube. 2/Turn the light on 3/Place slide on stage using stage clips 4/Position the lowest (shortest) objective into place 5/Without looking in the eyepiece, raise the stage to be very close to the objective but do not let them touch!
6
6/ Look through the eyepiece 7/Using the course adj., bring it into focus 7/ Use fine adj. to get into focus further (you may have to use the fine adj. only if in using highest obj.) 8. Move slide around if need – still keeping it clipped
7
Making a wet mount 1/Place specimen on slide 2/Place a drop of water on slide 3/Place cover slip on slide using the angle technique 4/Remove air bubbles by tapping gently with finger or pencil eraser 5/Dry excess water by taking a paper towel and dabbing the outside
8
To make a stained slide 1/Make a wet mount with water and coverslip 2/Place a drop of stain at one end of the cover slip 3/ hold a piece of paper towel at the other end of cover slip- this will draw the stain across the speciman
9
Calculating Magnification Eyepiece = 10x Scanning power obj = 4x Low obj = 10x Highest obj = 40x Ex. – eyepiece x low = 4x x 10x = 40x total
