Aim: qRT-PCR on polysomal and total RNA from NAc of mice after binge drinking
Methods : Polysomal RNA was extracted from IA 2BC 20% alcohol mice. After polysomal fractionation (according to Feng’s protocol), 500ng of RNA was used for the reverse transcription. Prosapip1 and GAPDH cDNA levels were tested by quantitative Real-Time PCR. Standard curves showed a slope between -3,0 and -3,6 and dissociation curves showed only one product amplified. Quantifications were made using the standard curve for the polysomal RNA and using the Ct values for the total RNA (not enough total RNA to do a calibration curve).
