“What is This Stuff?” Lab Prep INVESTIGATIVE QUESTION: How can we extract from cells the “stuff” that makes up chromosomes and carries the code of life? (please copy the IQ neatly into your lab notebook)

“What is this stuff?” Lab Prep please copy this list! MATERIALS: Cell smoothie – available at front demo table Woolite detergent – located on the black table Meat tenderizer – located on the black table Ethanol – in the freezer….please return ASAP

“What is This Stuff?” Lab Prep PROCEDURE Make a T-chart. Label the left column “procedural step” Label the right column “why step is necessary”

“What is This Stuff?” Lab Prep PROCEDURE, continued (As we conduct the lab procedure, think about WHY we do each step. Record your reasonable explanations in the “why step is necessary” column.) Step 1: blend 100 mL peas with 200mL H 2 O and a pinch of salt for 15 seconds Step 2: strain the solids so that you have a “pea smoothie” (do not drink!) Step 3: obtain 20mL of the smoothie in your beaker (no “why” is necessary for this step)

“What is This Stuff?” Lab Prep PROCEDURE, continued (As we conduct the lab procedure, think about WHY we do each step. Record your reasonable explanations in the “why step is necessary” column.) Step 4: add 3mL of Woolite detergent to the beaker; swirl gently. Step 5: wait 10 minutes, gently swirling the mixture every 2 minutes Step 6: Pour mixture into the test tube (no “why” is necessary for this step) Step 7: Use wood stick to add a pinch of meat tenderizer; mix gently

“What is This Stuff?” Lab Prep PROCEDURE, continued (As we conduct the lab procedure, think about WHY we do each step. Record your reasonable explanations in the “why step is necessary” column.) Step 8: SLOWLY Add a ¾” layer of ice cold ethanol to the test tube. DATA Soon, the “stuff” will become visible in the ethanol layer! Record what you see with a labeled sketch. Use color in your sketch.
