Cytomorphology of Normal Blood Cell Zhaoxindong

erythron pronormoblast Early normoblast:basophilic Intermediate normoblast: polychromatic Late normoblast:

pronormoblast Shape:round or oval 15-20um in diameter Nucleus: large, round or oval reddish purple in color granulous chromatin Pattern with 1-2 nucleoli Cytoplasm: deep blue and opaque Small amount, may show Smalltumorlike protrusion

Early normoblast:basophilic Shape:round or oval 10-18um in diameter Nucleus: large, round or oval chromatin pattern slightly coarser, nucleoli indistinct Cytoplasm: deep blue and opaque Small, perinuclear pale zone is obvious

Intermediate normoblast: polychromatic Shape:round or oval 8-15um in diameter Nucleus: smaller, round or oval chromatin more condensed no nucleoli Cytoplasm: blue-gray to pink-gray

Late normoblast: Shape: round 7-10um in diameter Nucleus: pyknotic nucleus Cytoplasm: Pinker than previous stage

Myelocyte series Myeloblast Promyelocyte Myelocyte Metamyelocyte Band neutrophil Segmented neutrophil

Eosinophlic granules the neutrophlic granules are fine and stain a blue-pink color with Wright stain. the neutrophlic granules are fine and stain a blue-pink color with Wright stain. are larger than Neutrophilic granules. These round and oval-shaped granules are orange and have a Glassy or semiopaque texture. basophilic granules have a dark blue-back color and dense appearance.

Myeloblast This cell has an average overall diameter of 10 to 20um,the nuclear chromatin is finely reticular, with 2 to 5 light-staining nucleoli. the cytoplasm is sky blue, small in amount and lacks granules.

Promyelocyte The cell is larger than the blast stage, with an average diameter of 12 to The nuclear chromatin is more condensed than in the blast, and nucleoli are present. the cytoplasm is a pale grayish blue. Cytoplasm have a few azurophilic granules.

Myelocyte The cell has an average diameter of 10 to 20um. The nucleus has a more oval appearance than in previous stages, no nucleoli, and the chromatin is much more clumped than in previous stages. Cytoplasm have a few neutrophilic granules.

METAMYLOCYTE The cell most characteristic feature is that the nucleus begins to assume an indented or kidney-bean shape. Which will continue to elongate as the cell matures through this phase. The chromatin continues to become more condensed or clumped.

Band granulocytes Band granulocytes has a typical elongated nucleus. The chromatin is very clumped. The cytoplasm have a fine and abundant specific pink neutrophilic granules.

The segmented granulocytes The cell has a characteristic multilobed nucleus, the separate lobes are attached to each other by a fine thread like filament.

megakaryocytic series Megakaryoblast Promegakaryocyte Granular megakaryocyte Platelet-producing megakaryocyte Nude nucleus megakaryocyte Platelet

Megakaryoblast The cell size can be vary from 15 um to over 50 um in diameter. it is a round cell The compact nucleus has an overall color of pink and the chromatin is a red- purple color, normally, megakaryoblasts have several nucleoli, the nucleus is surrounded by basophilic cytoplasm. cytoplasmic pseudopods are frequently seen. No granules.

Promegakaryocyte The cell size can be vary from 30 um to over 50 um in diameter, the chromatin continues to darken in color with increased clumping seen in the nuclear pattern, two or more distinct nucleoli may be seen. The quantity of cytoplasm increases; the basophilic color persist.pseudopods are frequently seen. Fine azurophilic granules are most abundant adjacent to the nucleus.

Granular megakaryocyte Megakaryocytes are the largest bone marrow cells with an average size of up to 100 um, no nucleoli distinctive feature of the megakaryocyte is that it is multilobular, not multinucleated,

Platelet-producing megakaryocyte on completion of maturation the fully mature lobes of the magakaryocyte shed platelets excplosively from the edges of the cytoplasm.

Nude nucleus megakaryocyte

Platelets Platelets have an average diameter of 2 to 4 um with younger platelets being large than older ones. platelets have no nucleus, the cytoplasm is light blue with evenly disperped, fine red- purple granules.

Plasmacyte series Plasmablast Proplasmacyte Plasmacyte

Plasmablast Shape:round and elliptical 14-18um in diameter Nucleus: round or oval and eccentric. reticular chromatin, nucleoli indistinct Cytoplasm: basphilic and opaque cytoplasm.abundant and vacuolate without granules

Proplasmacyte S:round and elliptical 12-16um in diameter N:round or oval and eccentric. chromatin coarser and more clumped than in the plasmablast stage, nucleoli invisible. C: basphilic, paranuclear clear zone may become visible, vacuolate, nongranular or a few

Plasmacyte S:round and elliptical 12-15um in diameter N: round or oval and eccentric with nucleoli wheel spoke chromatin pattern C:deeply basphilic, with vacuoles or frothy, nongranular
