Bio-bandage which injects medicine to organ directly using extension of the Hydrogel 서강대학교 화공생명공학과 연성물질공학연구실. 김정민 김영대 양태경 임승모 서강대학교 화공생명공학과 - 화공생명공학심화설계, 2014 Background Manufacture process Bandage for organ Invivo bandage! + injection drugs to organ directly + dissolve itself in body Bilayer structure Objective Previous study Experiment & Result Step1. Prepare Hydrogel Step2. 1 st Layered glass Step5. Chain-polymerization of 2 nd hydrogel Curvature of bilayer Hydrogel properties technogel,, plastic solution company Hydrogel Timoshenko equation Extension / Micro size processing Physical property / Micro size Step3. Photopolymeriztion of 1 st Hyrogel Step4. Stack 2 nd layered glass Drug is injected by adequate circumstance l l t1t1 t2t2 E1E1 E2E2 Network structure polymer Properties of hydrogel are similar with the cell Easy processing Drug release by specific Temperature, ionic/magnetic force, PH or wavelength ApplicationPilot test product 1 st gel chain 2 nd gel chain Step6. Remove glasses & residue Degassing Hydrogel Residue should be removed Control degree of crosslinking Control bilayer thickness Low Modulus hydrogel (Extension occur) High Modulus hydrogel (Forming hard structure) Remove gas in molecules by vacuum pressure Remove Oxygen Bio-bandageControl of injected materialsWhere can we use Bio-bandage? Control of injection time Advantage Change types of medicine Extension of hydrogel by liquids Possible to inject drug of your choice Possible to control the quantity of injected medicine Control of duration of medicine efficiency Controling the quantity of crosslink can change duration of drug emission Duration of medicine - Damaged organ - Place of sutures - Bleeding wound Attach Bio-bandage to damaged or wounded organ After attached, medicine continuously is injected by extension of hydrogel Hard structure that prohibit gel deformation or damaged 1 st hydrogel Similar properties with cell 2 nd hydrogel Inject medicine to organ by extension of hydrogel Low extension that prohibit gel expansion to wrong area Control of size or shape Swelling Swelling by Modulus Control of gel size easily Control of shape by Timoshenko equation Hydrogel Usually used bio-material Easy control of releasing Easy control of size No technology for drug delivery to organs Drug delivery Purpose Hydrogel bandage Similar properties with organ OctoDex, Octoplus company Cure surgical site We can control releasing time and types of drug Hydrogel has good bio-compability We can find hydrogel is appropriate for drug delivery So We attempt to develop bio- bandage for internal organs (a) Schematic degradation process (b) photographs of hydrogels in dry state and during degradation process We can make biodegradable hydrogel with natural polymers such as collagen, DNA Also, with degradable cross linking agent, we can synthesize biodegradable hydrogel This hydrogel is liquid in room temperature. So it is more convenient to use and more applicable material to medical field We can make bio-adhesive hydrogel synthesizing pluronic-thiol ( Plu-SH)and catechol-functionalized hyaluronic acid. It is designed to mimic the properties of mussel No needs to additional operation for removing bandage Bio-adhesive Transportation of drug Swelling Dried hydrogels have drugs - tight network structure A A B B Experiment of sample’s diffusionExperiment of diffusion rate 0min 15min 30min Being able to control sample diffusion rate using cross link ratio Bio-degradable Expected occupation of demands Swelled hydrogels release drugs - loose network structure Polymerization Photopolymerization Chainpolymerization 1. Drug can be transported directly without additional injecting medication. 2. We can control the site where drug is injected. 4. The method of uses is simple. the hydrogel bandage can be applied for biotechnology such as surgical operation, plastic surgery, bio-sensor, drug delivery, etc. Diffusion rate of sample Difference of cross link ratio Cross linkerMonomerWater(dye)Initiator Crosslinker ratio Diffused dye Diffusion Measure average intensity on rectangle of UV dye by ZEN software Detection of UV dye Measure intensity of UV dye Intensity is positively time dependent 3. We can control the effective duration of medicine
