Figure 3 Cytokine gene expression in PBMC stimulated with PPD or MBP in vitroCytokine messenger RNA transcripts were isolated from peripheral blood mononuclear cell (PBMC) cultures described in figure 2 and analyzed by quantitative real-time PCR (A–F). Cytokine gene expression in PBMC stimulated with PPD or MBP in vitroCytokine messenger RNA transcripts were isolated from peripheral blood mononuclear cell (PBMC) cultures described in figure 2 and analyzed by quantitative real-time PCR (A–F). The box-and-whisker plot for each visit represents the peak responses from 6 patients. High levels of interferon (IFN)-γ and tumor necrosis factor α (TNF-α) gene expression (A, C) correlated with high levels of secreted cytokines in response to purified protein derivative (PPD). Low transcript levels of IFN-γ (D) and TNF-α (F) in response to myelin basic protein (MBP) and interleukin (IL)-10 (B) in response to PPD reflected the low levels of cytokine secretion. Analysis of individual patient responses to either MBP or ATX-MS-1467 revealed that only 1 individual, expressing DRB1*15, transcribed IL-10 in response to both MBP and ATX-MS-1467 (G–I). Heather B.
