Figure 1 Schematic Representation of the Differential Effects of IL-12, IL-18, and TGF-β on Expression of IFN-γ by NK Cells IL-12 and IL-18 treatment downregulates the ability of NK cells to respond to TGF-β through downregulation of one chain of the TGF-β receptor and the TGF-β-activated transcription factors SMAD2 and SMAD3. In contrast, TGF-β downregulates IFN-γ expression though inhibition of the transcription factor T-bet and direct interaction of the SMAD proteins with the IFN-γ promoter.
