TGF-β signaling at its heterodimeric receptor (TGF-βRI–TGF-βRII) results in the phosphorylation of the TGF-β-regulated transcriptional activators Smad2 and Smad3 proteins, their association with Smad4, and translocation to the nucleus. Smad-dependent induction of Foxp3 in turn reinforces TGF-β-dependent gene expression, including the transcription of TGF-βRI.
