Fig 2 Involvement of TNF in immunity against mycobacterial infection and the effect of TNFis on mycobacterial infection. NO, Nitric oxide; T reg, regulatory T cell. 1, TNF recruits inflammatory cells with chemokines and activates macrophages. TNFis lead to increased bacterial burden and decreased chemokine levels in animal models.28 2, TNF and IFN-γ synergize to stimulate nitric oxide production in macrophages to kill mycobacteria.24 3, After infliximab treatment in an in vivo study, the number of CD8+ T cells expressing cytotoxic granules decreased, and the antimicrobial activity against mycobacteria decreased.29 4, TNF is essential for the formation and maintenance of granulomas. Blocking TNF disrupts granuloma structure and enables mycobacterial dissemination.28 Journal of Allergy and Clinical Immunology 2016 137, 19-27DOI: (10.1016/j.jaci.2015.10.023) Copyright © 2015 Terms and Conditions
