Fig 1 Role of AP-rPTP on IL-4 receptor (IL-4R), IL-13 receptor (IL-13R), and IL-23 receptor (IL-23R) signaling. Binding of cytokine to its receptor induces the recruitment of JAKs. JAKs are phosphorylated, either through autophosphorylation or phosphorylation by other kinases, which increases their kinase activity. JAKs phosphorylate tyrosine residues on the cytoplasmic tail of the receptor, allowing STAT recruitment (step not illustrated). STATs are then phosphorylated by JAKs. STAT phosphorylation results in dimerization, followed by nuclear translocation, during which they will regulate gene expression (steps not illustrated). AP-rPTP dephosphorylates various proteins, including JAK1, JAK3, STAT3, and STAT6, interfering with IL-4, IL-13, and IL-23 signaling at different levels.
