The Release Of Curcumin From Alginate Beads In Different Media Esra Aksoy1, Melisa Berberoglu1, Cagla Ergun2, Huceste Catalgil Giz1 1Istanbul Technical University, Department of Chemistry, Istanbul, Turkey 2Bogazici University, Department Of Chemical Eng, İstanbul, Turkey INTRODUCTION Alginic acid is a linear copolymer with homopolymeric blocks of D-mannuronic acid (M) and L-guluronic acid (G) residues, covalently linked together in different sequences or blocks [1]. Sodium alginate is the sodium salt of alginic acid. Its empirical formula is NaC6H7O6. Sodium alginate is a gum, extracted from the cell walls of brown algae [2]. Figure 1. It is used in drug and food applications due to its good biocompatibility, biodegradability, non-toxicity, gelation and film formation properties [3]. Soluble sodium alginate can be cross-linked using calcium chloride, forming insoluble calcium alginate particles with different diameters, depending on the concentrations of sodium alginate and calcium chloride. [4]. A bioactive compound can be protected from environmental destructive factors, and then can be solubilized and delivered in a controlled manner by encapsulation. [5]. Curcumin has been used as an orange food coloring for a very long time. Recently it has been found to have beneficial health effects like antioxidant, anti-inflammatory, antimicrobial and anticarcinogenic activities and has gained importance. Its insolubility in water is the main obstacle in producing useful drugs, which take advantage of these properties. In order to increase the applicability of curcumin in food and drug systems its solubility in water has to be increased. CONCLUSION Figure 2. Chemical structure of curcumin (keto-enol forms) The aim of this work was to prepare curcumin loaded alginate beads and observe release rates in different media. As a release medium, water, Tween80, simulated gastric fluid (SGF) and simulated intestinal fluid (SIF) were chosen. Complexes were encapsulated in alginate beads and release rates were monitored by online monitoring of polymerization (ACOMP) system with an UV detector. Release profiles in Water and Tween 80 were monitored by ACOMP system and a data point was recorded at every second and monitoring was continued for 10-20 hours. Beads in tween80 retained their shape and continued to release for long periods, however in water, beads did not show a long release period. In water only the 0.1 % medium viscosity alginate released the highest amount of curcumin for a long time period. Release profiles in SGF and SIF monitored by sequential sampling show that the 0.1 % low viscosity alginate releases the highest amount of curcumin. Beads were stable in SGF, however they disintegrate in SIF at the end of the period. In all release profiles except in SIF, the dilute solution of alginate (0.1%) releases more curcumin than the concentrated solution of alginate (0.5%). The results show that in the dilute solutions the alginate polymer chains are broken down more by Ultrasound and so they can bind more curcumin. Stress-strain experiments have shown that, beads prepared from low viscosity alginate are softer, having higher strain values than the medium viscosity alginate. METHODS In this work Curcumin saturated low and medium viscosity sodium alginate solutions were prepared at different concentrations (0.1% and 0.5%) in ultrasound medium (80Watt, 35kHz) at 25°C. Dilute curcumin-alginate solutions were concentrated by vacuum distillation for bead formation. Curcumin saturated alginate solutions were dripped into 2% (w/w) CaCl2 solution and curcumin containing alginate beads were prepared. The release profiles of all beads prepared from 0.1% and 0.5% low viscosity alginate and from 0.1% and 0.5% medium viscosity alginate were monitored by ACOMP system in water, and Tween80. Release profiles in SGF and SIF were monitored by sequential sampling. Bulk modulus of beads were measured by stress strain measurements. REFERENCES Jejurikar A., Lawrie G., Martin D. et al.
