4 In general… Higher enzyme concentration (up to a certain point) = faster reaction rate Higher substrate concentration (up to a certain point) = faster reaction rate Too high/too low temp = slower reaction rate Too high/too low pH = slower reaction rate With inhibitors= slower reaction rate

5 Must Know/Beautiful things about enzymes They can be used over and over- 1000’s of substrate molecules can be changed per second… They are very specific meaning they are an expert at only one reaction. Won’t do anything else. They do not perform magic – they don’t change the rules of the reaction (equilibrium or delta G) – we just get there faster.

6 Agenda 10/10 Finish/Review Enzymes (10-15 min) Toothpickase Lab – I’ll walk you through first 2 counts, then you do on your own –Run for a total of 4 minutes (After step 13, count for 120 more seconds) –Share data and go over discussion questions –Start graph if can Homework- Ch. 8 as before (notes/concept checks due tomorrow) –Print out and read Lab 2 Enzyme Catalysis from website (Cells page) - Prelab – Identify control, independent variable etc. for part A and B and know procedure!!! Then answer questions 1-6 at end. –Toothpickase worksheet with graph due tomorrow

8 Agenda 10/11 Quick go over Toothpickase lab rate calculations, graph, and questions (5 min) – per. 5 feedback inhibition Toothpickase lab and Ch. 8 notes – leave out for me to check during lab Quick go over technical details of setup and control, indep. and dep. variable, constants, hypothesis http://www.explorebiology.com/apbiology/labs/lab08.html Run part A and B – do 5 minute runs (for B – some groups start with 0.25 and some with 0.75 so can share data if run out of time) Try to graph as you go – part A and B go on same graph with lines labeled, answer 7a-c Pick parts C,D,E or F for tomorrow – do quick prelab with your group Homework –No homework night – nothing due tomorrow –Will do in class - Prelab – Hypothesis and Identify control, independent variable etc. for part you are assigned for tomorrow and know procedure!!! –Lab packet will be due Tuesday with data tables and graphs attached –Rest of Ch. 8 online assignment due next Mon. 8 am Enzyme Quiz next Tuesday

9 Agenda 10/12 Run parts C, D, E or F – I’ll check in and be sure you know what you’re doing During your trials – get concentration data from other groups to complete part B -- graph A and B on one side (4 labeled lines) - graph your other part on the back (4 labeled lines – graph control again) - answer questions for your part and part A and B Homework – –Rest of Ch. 8 online assignment due Mon. 8 am –Finish graphing as needed for your part –Try to answer as many questions as you can – especially for your part and #12 –Completed lab packet due Tuesday Enzyme Quiz on Tuesday HAVE FUN AT HOMECOMING! 
