3 | What is a spot test? A spot test is … a simple chemical reaction that allows a substance to be identified. HeroinCocaine In drug testing… a drug or class of drugs gives a characteristic visible change when mixed with another chemical (the reagent) a sample, such as urine, is tested with the reagent the presence of the drug (or class of drug) is evidenced by the characteristic visible change; this is a positive test if the characteristic change is not seen, it is a negative test
