In the first set of experiment detailed in Fig. 1 <$REFLINK> , at the termination of the study, plasma concentration of human IGFBP-3 was estimated using quantikine ELISA, as mentioned in “Materials and Methods.” A, standard curve using recombinant human IGFBP-3 to extrapolate the plasma level of human IGFBP-3. Absorbance of the developed color in reaction was determined using a microplate reader set to 450 nm with correction wavelength at 540 nm, and a standard curve was established with known concentrations of IGFBP-3 at logarithmic scale. B, IGFBP-3 concentrations (ng/ml) in plasma samples were calculated by four-parameter logistic curve-fit generated standard curve and are represented as mean of 7–8 plasma samples from individual mouse in each treatment group. ∗, P < 0.05; bars, ± SE.
