Expression of active Rho partially rescues Erk1/2 activation and peripheral FA phenotype in RIAM-knockdown cells. (A) Mock and D11 cells were attached for the times indicated on FN, and subsequently subjected to Rho GTPase assays. (B) Cells were transfected with RhoA wt-GFP, RhoA CA-GFP or GFP vectors, and transfectants were subjected to immunoblotting using anti-GFP (top), or vinculin, phospho-Erk1/2 or total Erk1/2 antibodies. (C) The same transfectants attached to FN were analysed by confocal microscopy using anti-paxillin antibodies. Shown are representative cells from each transfectant condition (n>100 cells).
