Fig 1 Proportions of NOTCH1+ and NOTCH2+ cells are increased in various T-cell compartments in peripheral blood from patients with asthma. A-B, Flow cytometric gating of CD3+CD4+ TH cells (excluded for CD127−CD25hi regulatory cells), CD45RA− memory T cells, and CD45RA−CXCR5−CCR6−CCR4+CXCR3− TH2 cells (Fig 1, A). B, An example of NOTCH1 and NOTCH2 expression on T cells. C, Proportions of NOTCH1+/NOTCH2+ CD4− T cells, CD4+ T cells, naive T cells (CD45RA+), memory T cells (CD45RA−), and TH2 cells (CD45RA−CXCR5−CCR6−CCR4+CXCR3−). D, Correlations of proportions of NOTCH1+/NOTCH2+ cells within naive T cells, memory T cells, and TH2 cells, measured in peripheral blood from patients with asthma. E, ACQ scores of patients with high (above median) or low (below median) proportions of NOTCH1+/NOTCH2+ TH2 cells. F, Mean fluorescent intensity of CD25 and CRTH2 on NOTCH1+ and NOTCH1− memory CD4+ T cells. Data were acquired with an LSR II flow cytometer operated using FACS Diva software 6.1 (BD Biosciences, Erembodegem, Belgium) and analyzed using FlowJo (Tree Star Inc, Ashland, Ore). Data are shown as symbols for individual patients/controls, together with horizontal bars for median values. Comparisons between groups (Fig 1, C, E, F) were evaluated using a Mann-Whitney U test. P values were 2-sided and analyses were performed using GraphPad Prism software (version 5.01). P < .05 was considered statistically significant. CRTH2, Chemoattractant receptor-homologous molecule expressed on TH2 cells; FSC-A, forward scatter-area. *P < .05, **P < .01, ***P < .001. Correlations (Fig 1, D) were analyzed using a Pearson correlation test.
