FIG. 1 Schematic representation of cis- and trans-splicing. The diagram depicts a four-exon gene (1, 2, 3, and 4). The primary RNA transcript, pre-mRNA, containing both intron and exon sequences is shown. RNA cis-splicing (left arrow) will generate mRNA molecules containing four exons, from which three introns are spliced; in RNA trans-splicing (right arrow), a pre-trans-splicing molecule (PTM), which carries an exon X, is linked through base-pairing to the third intron. The 5′ splice donor of this intron is then joined to the 3′ splice acceptor in the PTM by spliceosome-mediated trans-splicing, which generates an altered mRNA in the reprogrammed transcript.
