2 Figure 1 Effect of Exogenous GABA on Metabolism and Gene Expression under C-Limiting Conditions. (A) Experiment set up. Metabolite extraction of seedlings (100mg) and the elaboration of the data followed an established protocol (Lisec et al., 2006). RNA extraction was carried out on seedlings from the different treatments with and without 1mM GABA (based on metabolite profiling), using TRIzol (Invitrogen) protocol. Microarray analysis was performed according to the standard Affymetrix protocols ( accessed 2 March 2014) on ATH1 Array chip. Differentially expressed genes were chosen according to FDR < 0.05 and fold change > 4. Functional annotation, overrepresentation analysis, and metabolic pathways display were performed using the DAVID database ( accessed 2 March 2014) and the KEGG database ( accessed 2 March 2014). (B) An effect of exogenous GABA under FN (green bars) and low-C (blue bars) conditions on a relative content of the primary metabolites (y-axis). Bars represent average value ± SD (n = 4). Asterisks represent significant changes with respect to GABA addition within each condition (t-test p < 0.05). (C) An effect of exogenous GABA on gene expression level under low-C conditions. Only significant changes according to p-value, fold change, and enrichment score are presented. (D) An effect of exogenous GABA on gene expression level associated with a metabolism of N-compounds.
