3. What is TVC? TVC = Total Viable Count HCC, HPC, TCC Indication of the size of the microbial community in a water system Microbes will flourish with no disinfection measure present ie Chlorine 4187 Guideline Presentation- March 2015

Pour plate – 1ml inoculation – growth medium added and incubated at a known temperature for 2-3 days. Potable =22 and 36 deg C respectively Membrane Filtration (MF) – 100ml inoculation - growth medium added and incubated at a temperature between 30 – 35 deg C for 5 days 4187 Guideline Presentation- March 2015

9 4. What are Endotoxins & How are they analysed?
Endotoxins are large molecules consisting of a lipid and a polysaccharide (LPS) and are found in the outer membrane of Gram-negative bacteria Endotoxins are often released upon cell death but can be excreted by gram –ve bacteria as a defence mechanism Endotoxins when introduced into human body, can cause a fever-like reaction 4187 Guideline Presentation- March 2015

10 4. What are Endotoxins & How are they analysed?
ALS currently perform Bacterial Endotoxin analysis in our Brisbane laboratory Based on clotting reaction of horseshoe crab blood Sample is incubated with an LAL standard (Limulus Amebocyte Lysate) at a known sensitivity and the clotting time is measured and plotted on a standard curve The sample is collected in a sterile & non-pyrogenic 50mL tube HT of 24hrs – currently running extended HT data TAT 5-7 days – Fast TAT available (1 day)

11 5. Sampling & Transport to the Laboratory
Aseptically collected – flaming / ethanol Appropriate containers – sterile, non-pyrogenic Correct chilling - <4 deg on receipt Holding time – TVC analysed within 24hrs of collection
