Agenda 4/24 Recombinant DNA warm up Gel Electrophoresis Techniques
Biotech Review Questions Homework PCR Video and Notes
2
Recombinant DNA Steps From memory, attempt to write out the steps of recombinant DNA using a specific example
3
Solve the crime…
4
Polymerase Chain Reaction (PCR)
When a DNA sample is found, it must be larger enough to work with. PCR is used to amplify the sample of DNA into a big enough sample to analysis using gel electrophoresis

PCR
6
Restriction Enzymes Restriction enzymes cut at specific sequence in DNA They are used in gel electrophoresis to create unique ‘cuts’ for each person These smaller pieces of DNA are then loaded into a gel to be analyzed There are also used in recombinant DNA to cut out the gene of interest

Restriction Enzymes
8
Gel Electrophoresis Overview
9
Gel Electrophoresis Smaller pieces of DNA travel farther
A ‘comparison’ DNA sample is used Can be used to determine size Or to match an unknown sample DNA travels from the negative side (anode) to the positive side (cathode) What does that mean about the charge of DNA?
