1 MMP-7 and TIMP-1, New Targets in Predicting Poor Wound Healing in Apical Periodontitis

2 Figure 1 Individual MMP and TIMP gene mRNA expression levels in CAA and AAP in comparison to control samples. Results show mean cycle threshold (Ct) values from triplicate measurements of the target gene with normalization to the internal control beta-actin. Relative changes in gene expression were calculated using the delta delta Ct method (18). The expression of all MMPs and TIMPs genes was significantly higher in both CAA and AAP cases when compared with controls (P < .001) (top panel). MMP-2, MMP-7, and MMP-9 mRNA levels were higher in CAA cases (bottom left), whereas TIMP-1 mRNA was higher in AAP cases (bottom right) (P < .05). 3 Figure 2 Results of immunohistochemistry analysis for MMP-7 and TIMP-1 proteins in tissue samples of AAP and CAA cases (magnification 40× [A and B] and 10× [C and D]). Journal of Endodontics  , DOI: ( /j.joen ) Copyright © 2013 American Association of Endodontists Terms and Conditions

4 Figure 3 Immunocytochemistry analysis of MMP-7 and TIMP-1 proteins in macrophages (A) without LPS treatment (controls) and (B) after treatment with LPS. The expression of MMP-7 and TIMP-1 appears increased in LPS-infected cells compared with noninfected cells. (C and D) MMP-7 and TIMP-1 colocalize in the Golgi apparatus, and (D) their expression appears increased in LPS-treated cells (magnification 400× [A and B] and 1,000× [C and D]).
