1 Wednesday Agenda: Pre-Lab Quiz
Lab Day 1 Procedures – Incubation Temperature Difference HW: Day 1 Post Lab, Read for Day 2 Wednesday

2 Thursday Agenda: STEM Warm-Up Notes: Cloning 20.3 Lab Day 2 Procedures
Lab Write-Up Due March 4th Thursday

3 Thursday Agenda: Warm-Up Notes: Cloning 20.3
List of BioTech Cons: BioTech Agriculture, Medical, and Industrial Gel Electrophoresis Notes Lab Write-Up Due March 4th Thursday

4 Order for the remainder of the lab:
Post Lab Questions Day 1 – Last night Day 2 Equations and Pictures Day 2 Post Lab Questions Conclusion: Summarize, Error, Apply

5 CLoning Plants – Totipotent Cells – Can give rise to all cells of the plant (Cutting) Animals – nuclear potential to differentiate is restricted as the organism ages Many different types of animals have been cloned since Dolly. What are some conclusions that have been drawn from cloning of animals? They don’t always look and act identical to their donor. Environmental impacts and random phenomena – mostly due to epigenetic changes of chromatin.

6 Cloning and Stem Cells Where do they come from? Which type is better? Possible therapeutic uses? Latest: Learned to deprogram differentiated cells and then reprogram them

8 Rat Heart

9

10 BT Corn Bacillus thuringiensis and European Corn Borer
Bt protein ingested Binds to the gut wall, breaks down Insect dies of septicemia as bacteria multiply in the blood Kills beneficial insects Humans?

11 Problems with GMOs

12 Around the world 38 countries that have banned GMOs – unknown risks
Some countries have limits - # hectares Rules on importation, trade arguments Many GM Labeling Required

13

14 Lab Discussion What are some factors that are essential for bacteria to grow? Inhibit growth?

15 Lab Discussion and predictions
LB - Plasmid LB + Plasmid LB/AMP – Plasmid LB/AMP + Plasmid

16 Friday STEM Agenda: G.M. O? Create GMO Con List
Gel Electrophoresis Notes Socrative Quiz HW: PreLab 9 Agenda: Socrative Quiz Lab Discussion Finish Lab 8 Friday

17 Order for the remainder of the lab:
Post Lab Questions Day 1 – Last night Day 2 Equations and Pictures Day 2 Post Lab Questions Conclusion: Summarize, Error, Apply

18 STRs (Short tandem Repeats)
FBI: DNA: 1988 Highly variable areas of our DNA, person to person (2 to 5 nucleotides) Vary at same locus on different homologs ACTA 30 times ACTA 15 times 30, 15 Number of repeats can be identified using Gel Electrophoresis

19 Gel electrophoresis What is it? Genetics Utah Animation

20 Standard HindIII Lambda DNA Sample
Ladder or marker is run to show known fragments for comparison.

21 Standard Curve (plot of distance migrated vs. DNA size (base pairs)

22 Practice A bloody crime scene has occurred, and police have collected several blood samples. The following table shows the number of repeats for three STR markers from those samples. (Normally there are 13 STR markers) Analyze the data and determine which suspect you would charge with the crime. Source of Sample STR Marker 1 STR Marker 2 STR Marker 3 Blood at Crime Scene 12, 15, 18, 19 9, 14, 17 10, 15, 16, 20 Victim 15, 19 9, 17 15, 20 Suspect 1 12, 21 15, 17 10, 18 Suspect 2 12, 18 14, 17 10, 16

24 Lab 9 Restriction Enzymes and DNA
Problem: How can DNA fragments be separated and analyzed by gel electrophoresis? Restriction enzymes (RE’s): recognize specific DNA sequences and cut DNA at these sites RE’s in this lab: EcoRI and HindIII EcoRI: sequence GAATTC, cuts G | AATTC HindIII: sequence AAGCTT, cuts A | AGCTT RE’s cut DNA so that a gel electrophoresis can be run to create a DNA fingerprint

25 Friday Homework: Lab 9 PreLab: PreLab Quiz Monday (Set up and safety)
Introduction: One-two paragraphs of key terms Pre-Lab Questions 1-5 List Materials Day One and Day Two Safety SAFETY S A F E T Y Day 1 Procedures (Five activities to read over) PreLab Quiz Monday (Set up and safety) Lab 8 and 9 Due: March 4th
