4 Essential Question How do we prepare bacteria for viewing? Standard 11C – summarize the role of microorganisms in both maintaining and disrupting the health of both organisms and ecosystems. PG 113

5 Culturing Bacteria Lab Q1: Label Petri Dish with location and the circle found on your handout Q2: Collect your Petri Dish, draw and color your petri dish EXACTLY as it looks on your handout. Q3: Answer after lab, use your notes Q4: Create up to 2 wet mounts from DIFFERENT bacterial locations on your petri dish. (1 at a time) -Put ONE drop of water on the center the slide - Use a toothpick to pick up some bacteria from the first location on your petri dish. -Stir the end of the toothpick in the drop of water and make a VERY THIN smear on your slide. Throw the toothpick away in the designated beaker. -Bring your wet mount to me so I can stain (crystal violet dye) the slide for you. -Place the cover slip on top of the stain/smear. Place down diagonally. -Use a paper towel on one end and add a drop of water to the other to drain out (OSMOSIS) the purple color over the sink. -Your Wet Mount is complete! -View under the microscope using the lowest objective, and move up. -Draw and color what you see on the handout. EXTRA CREDIT! Take a picture of your bacteria slide through the microscope and email it to me Enrique.Garza@birdvilleschools.net Answer questions with your shoulder partner only.
