Results vs Interpretation Results: What is observed with your eyes Interpretation: What the observations and results mean

Pure vs mixed Pure: It has only one type of organism Mixed: It has 2 or more kinds of organisms Contamination: Presence of unwanted organism

SUBCULTURE Removing one isolated colony from its original plate and transferring it to a new plate to create a pure culture from a mixed culture. It is important to identify unknown organism.

Dilution technique Example- Pour plate, Streak Plate and Spread plate Used to separate and isolate different organisms from one another by decreasing the concentration of original sample and to obtain a pure culture via subculturing

BROTH GROWTH PATTERNS

DEEP GROWTH PATTERNS

GROWTH PATTERN IN SLANT

OBSERVING PLATES in locations where bacteria are crowded closely together, confluent growth appears. In areas where the bacteria are more spread out individual colonies should appear. Each colony is the result of a single bacterial cell undergoing multiple divisions during the period of incubation. If your plate streaking was done correctly, the initial streaking sector should contain a fairly confluent lawn of bacteria. The third and fourth sectors should contain individual colonies.

COLONY  Bacteria form colonies when grown in a solid medium. Bacteria  The colony is actually (and ideally) a group of bacterial clones since they are all derived from only a single parent cell.

COLONY MORPHOLOGY

How to define colony? White Opaque Margin- Undulate Whole-colony appearance-Rhizoid Elevation-Raised

Before Starting Wash your hands. Disinfect the working area Wear your lab coats

After finishing Those who could not get proper results should repeat the experiment in open lab Please fill out the media request form Remove all tape and place it in Waste bin Place the test tubes in discard area Petri-plates will go in biohazard bin Keep the lab coat back in drawer Submit your lab note-books
