Year 10 – Genetics and Biotechnology (Biology) Portfolio 3: Extracting DNA GET OUT YOUR MAC’S – NO PHOTOS NO GAMES NO MOANING!!!! THIS NEEDS TO BE FINISHED TODAY!

Year 10 – Genetics and Biotechnology (Biology) Know where genes and DNA are located Extract DNA safely from peas Evaluate experiment and answer questions relating to DNA stucture By the end of this lesson we should be able to:

Year 10 – Genetics and Biotechnology (Biology) Portfolio 3: Extracting DNA Aim: In this experiment I am trying to extract DNA from split peas Equipment List: –Blender –Peas –Alcohol (Ethanol) –2 test tubes –Test tube holder –Enzymes –Detergent –Pipette

Year 10 – Genetics and Biotechnology (Biology) Portfolio 3: Extracting DNA Methodology 1. blend 1/3 peas with 2/3 water for 3 minutes 2. Strain/Sieve ………………………. 3.Place ………………………2 test tubes …. 4.Add ………………………………detergent 5. ………………….pinch of enzymes to…………………….. 6. tilt …………………………… add 7. observe …… Observations: what did you observe? where was the DNA? Why was it in this layer? Insert a picture (full sentences)

Year 10 – Genetics and Biotechnology (Biology) Observations: Which layer???? Two separate layers are formed, Alcohol is less dense than water so it floats on top. The protein and grease parts find the bottom, watery layer the most comfortable place, while the DNA prefers the top, alcohol layer. DNA is a long string molecule that likes to clump together.

Year 10 – Genetics and Biotechnology (Biology) Portfolio 3: Extracting DNA Evaluation did you manage to extract all the DNA? Why/why not? Where there any sources of error? how could this experiment be improved? Questions: (Research: Google) Where do you find DNA in the cell? Do all cells contain DNA? What is DNA made up from? (think about the model we made) What are the 4 base pairs – how do they join together? What kind of people have the same DNA?

Year 10 – Genetics and Biotechnology (Biology) Know where genes and DNA are located Extract DNA safely from peas Evaluate experiment and answer questions relating to DNA stucture By the end of this lesson we should be able to:

Year 10 – Genetics and Biotechnology (Biology) Make a note of this info Draw a diagram or picture Discuss Think Investigate Watch Answer question
