Figure 1 Trafficking and Regulation of L-Type Calcium Channels (Left) The calcium channel α1 subunit is coassembled with β and α2-δ subunits in the ER and targeted to the plasma membrane where it associates with calmodulin (CaM). (Middle) Entry of calcium (yellow circles) via L-type channels in response to brief depolarization results in calcium binding to calmodulin, and feedback inhibition via calcium-dependent channel inactivation. Calcium-bound calmodulin can also signal to the nucleus to mediate gene activation. (Right) Prolonged calcium channel activity triggers reversible, calcium-dependent internalization into endosomes (E) due to calcium-dependent recruitment of eIF3e to the channel's II-III linker region. Possible transfer to a lysosomal compartment (Ly) may degrade the channel protein, leaving the L-type channel C terminus available for nuclear signaling.
