Figure 1 Psoriasin immunoreactivity is enhanced in lesional atopic dermatitis skin. (a) To localize the expression of psoriasin in human skin biopsies of AD patients and normal controls immunohistochemical staining was performed. Lesional skin of AD patients revealed increased psoriasin expression compared to nonlesional skin and healthy controls at correlating sites. Bars indicate 50μm. (b) Specificity testing was performed by preincubation of the antibody with various concentrations of natural skin-derived psoriasin. Blocking the antibody with higher concentrations than 0.06μg psoriasin resulted in complete inhibition of immunoreactivity in a tissue sample derived from the face of a healthy person. A defined area (0.55cm2) of the skin of AD patients and matching normal controls was rinsed with sodium-phosphate buffer (pH 7.4). Soluble psoriasin concentrations were determined using a psoriasin-specific ELISA with two mAbs generated against natural psoriasin. Each sample was tested in triplicates.
