Figure 1 PIDD Integrates Pro- and Antiapoptotic Signaling Pathways PIDD forms a cytoplasmic “PIDDosome” composed of PIDD, RAIDD, and procaspase-2 to promote apoptosis through a mitochondrial death pathway in response to genotoxic stimuli. PIDD also forms a nuclear PIDDosome containing RIP1 and NEMO (also known as IKKγ) to promote SUMO-1 attachment on NEMO in response to genotoxic and perhaps other stresses. Nuclear RIP1 is also posttranslationally modified in an undefined manner (denoted by “?”). Following NEMO sumoylation, ATM, also activated by genotoxic agents through the induction of DNA damage, such as double-strand breaks, stimulates a subsequent NEMO modification cascade. These modifications cause NEMO translocation from the nucleus to the cytoplasm, where it causes the displacement of IκB (which gets degraded). This leads to the release of free NF-κB in the cytoplasm, which then enters the nucleus to induce prosurvival gene transcription.
