Figure 1 TGFβ-Mediated EMT and Apoptosis (Left) TGFβ causes EMT and apoptosis in SMAD4-positive tumor cells by induction of EMT-associated transcription factors like SNAIL, which causes repression of another transcription factor KLF5. In the absence of KLF5, SOX4 causes transcription of pro-apoptotic genes like Bim and Bmf, leading to apoptosis. (Right) In SMAD4-negative cells, TGFβ signaling mediated through SMAD2/3 induces SOX4 induction, which along with KLF5 transcribes a pro-tumorigenic, epithelial progenitor gene expression program.
