Observatoire Océanologique de Villefranche sur Mer, BP 28, 06234 Villefranche sur Mer, France Long term zooplankton monitoring with the ZooScan -Insights from a 8 years project at the Observatory of Villefranche sur Mer and - Perspectives for a network of observation of whole plankton

Point B zooplankton time series is one of the longest and most complete in Europe TS1: 1966-2011, Regent Net (680µm mesh), oblique hauls 0-75 m, daily sampling TS2: 1966-2003, Juday Bogorov net (330 μm mesh), vertical hauls 0-75 m, daily sampling TS3: 1995 – 2006, WP2 net (200µm mesh), vertical hauls 0-75 m, weekly sampling TS4: 2003-2011, WP2 net (200µm mesh), vertical hauls 0-75 m, daily sampling  too many samples to analyse in a traditionnal way More than 20 000 net samples but pooled in weekly jars (>5000 jars).

 Partially analysed for target species until 1993  Degradation of the samples (formaldehyd leackage) State of the collection in early 2004 RADEZOO service proposed 3 complementary solutions for long term preservation and analysis. - digitalization - re-conditionning - semi-automatic analysis

RADEZOO Service : Digitalization and analysis of current and past zooplankton samples (since 1966) 10 WP2 5 Regent 10 JB 3 samples/week and more than 5000 in collection Digitalization Digital Archives Web site http://www.obs-vlfr.fr/Rade/Radezoo. Databases: NODC/IODC Web site http://www.obs-vlfr.fr/Rade/Radezoo. Databases: NODC/IODC Archiving the jars (bat Jean Maetz) Archiving the jars (bat Jean Maetz) Image analysis Publication

RADEZOO Service : Semi automatic analysis SAMPLES WP2 Time Series TRAINING SET Defining groups 1 2 Prediction by the model (Random Forest) Accuracy (confusion matrix) Cross-validation 3 (confusion matrix) PREDICTION in 15 groups Manual validation in 35 taxa

RADEZOO Service : Quasi Real time monitoring of zooplankton

New facility for plankton storage At OOV more than 25 000 zooplnkton samples (formaldehyde) and space for more. More space available (100m 2 ) if necessary. Scanning and barcoding so as to link images (web accessible) with environmental parameters and sample location in the new facility. Contact: stemmann@obs-vlfr.fr Before Now

RADEZOO Service : Perspectives End to end ecosystem monitoring analysis Toward an integrated network

Imaging systems, GREAT PERSPECTIVES Provide Indicators of ecosystem status (abundance, biomass, taxa, size spectra) Can be obtained using lab and in situ instruments, they provide high frequency data and data quality and harmonization can be achieved. These indicators can be used to develop mathematical models for systems where size is important (interactions between organisms and flux) Key of success o AGREED PROCEDURES (image format, treatment, semi-automatic recognition, intercalibration) o AGREED DATA MANAGEMENT and DISTRIBUTION o AGREED MODELING FRAMEWORKS o SUMMER SCHOOLS FOR THE USERS

DELIVERABLE: Common software for image analysis and data management for Flowcam, Zooscan, UVP and microscopy. We try to take into account other instruments (for other size ranges) DELIVERABLE: Common software for image analysis and data management for Flowcam, Zooscan, UVP and microscopy. We try to take into account other instruments (for other size ranges) 1: Fowcytometer2: FlowCam 3: Zooscan Key players of the « end to end » plankton can be monitored using semi-automated imaging systems UVP, VPR 4

Romagnan et al., 2012 OPERATION PILOTE POINT B: started in 2011 and now under publication

Image data base from FLOWCAM and ZOOSCAN exists and is used for image developing image analysis software OPERATION PILOTE POINT B: started in 2011 and now under publication

Merging data from the different imaging systems works but requires adaptation for the software (in progress, should be finished early 2013)

We try to take into account future development of future, in situ systems (cytometer, flowcam and UVP) 1-10µm >100 µm 10-100µm
