Peter Werner Final portfolio May 30, 2008

Zoom project

Zoom images: Morchella elata (morel) asci Zeiss Axioskop, Zeiss Axiocam CCD color images

More zoom images: Morchella elata (morel) asci Zeiss Axioskop, Zeiss Axiocam CCD color images

Story project: Mushroom microanatomy

Asci, Morchella elata (morel), 40x objective, phase contrast Zeiss Axioskop, Zeiss Axiocam CCD image

Asci, Morchella elata (morel), 40x objective, dark-field Zeiss Axioskop, Zeiss Axiocam CCD image

Asci, Morchella elata (morel), 10x objective, fluorescence, green (Rhod) ex Zeiss Axioskop, Zeiss Axiocam CCD color image

Basidium and chrysocystidia, Stropharia rugosoannulata (garden giant mushroom), 63x objective, bright-field Nikon fluorescence scope, Nikon microscope CCD/Nikon Elements composite color image

Basidium and chrysocystidia, Stropharia rugosoannulata (garden giant mushroom), 100x objective, fluorescence, green (TRITC) ex Nikon fluorescence scope, Nikon microscope CCD/Nikon Elements monochromatic image with color overlay

Cheilocystidia, Clitocybe saeva (field blewitt), 63x objective, bright- field Nikon fluorescence scope, Nikon microscope CCD/Nikon Elements composite color image

Various basidiospores: Chroomgompus vinicolor (upper left), 100x obj; Lactarius rubidus (upper right), amyloid (iodine blue) reaction with Melzer’s reagent, 63x obj; Panaeolina foenisecii (bottom), 100x obj; all bright-field. Nikon fluorescence scope, Nikon microscope CCD/Nikon Elements composite color images

Resolution vs Contrast: Diatom test

Overview: Gyrosigma balticum (diatom), 63x/0.80 NA objective, bright-field Test of resolution of surface frustules on Gyrosigma Slide by Klaus Kemp Zeiss Axioskop, Zeiss Axiocam CCD black and white images

Resolution and contrast: NA = 0.9 (condenser aperture wide open) Image well-resolved, but somewhat lacking in contrast

Resolution and contrast: NA = 0.8 (resoution of condenser matches that of objective) Image well-resolved and with good contrast

Resolution and contrast: NA = 0.4 (condenser diaphragm at 50% field of view in back focal plane) Image poorly resolved – Airy disk size is such that individual frustules blend together and give the false image of ridges on diatom surface. (“Bad” image)

Resolution and contrast: NA = 0.05 (condenser diaphragm stopped down to smallest aperture) Image very poorly resolved – Airy disks so large that many points in image run together and there is little resolution. (“Bad” image)

Plants

Leaf cross section, Dianthus, 20x objective, phase contrast Pre-prepared slide Olympus CKX41/31

Anther cross section, Lillium (lily), 4x objective, fluorescence, green (TRITC) ex, red em Pre-prepared slide Olympus CKX41/31

Stem cross section, Zea mays (corn), 20x objective, fluorescence, green (TRITC) ex, red em Pre-prepared slide Olympus CKX41/31

Pollen, Pinus (pine), 40x objective, fluorescence, green (TRITC) ex, red em Pre-prepared slide Olympus CKX41/31

Sporangium, Polystichum munitum (sword fern), 10x objective, fluorescence, blue (FITC) ex Olympus CKX41/31, Nikon Coolpix 4500 digital camera color image, through ocular

Insects and more diatoms

Aphid (unidentified species), 4x, dark-field (Ph3 ring substituted for PhL ring) Olympus CKX41/31 (Semi-”bad” image: air bubble and schmutz on otherwise good image)

Antenna, aphid (unidentified species), 40x, fluorescence, blue (FITC) ex Olympus CKX41/31

Leg, aphid (unidentified species), 20x, phase contrast Olympus CKX41/31

Diatom test slide, 20X, bright-field Slide made by master diatom artist, Klaus Kemp Olympus CKX41/31

Pleurosigma angulatum (diatom), from diatom test slide, 20x, phase contrast Olympus CKX41/31

Unknowns

Extra credit and other overlapping stuff

Make your own slide: all slides were made by me unless otherwise noted. This would constiute at least half the slides presented here. Dark-field: Several, including two darkfield images of morel asci (in both Zoom and Story projects) and one of the aphid images. “Bad” images: two of the diatom slides. Problems with the darkfield aphid slide are also noted.
