Oxidative stress, cause of aging and disease! April 21/2016 ATCO

My day job

ROS or free radicals

How OS affects me?

Heart disease – New studies show cholesterol isn’t an indicator or heart disease risk – it’s oxidation and inflammation Here’s why: Free radicals oxidize LDL (“bad”) cholesterol Oxidized LDL is very reactive and attacks muscle cells in artery walls The body responds to these attacks by sending out inflammation This triggers a series of reactions leading to atherosclerotic plaque This starts the free radical/inflammation cycle over again, building up more plaques Plaques break off, leading to heart attacks and strokes

Arthritis–Oxidative stress ages the cartilage faster and increases inflammation, which also spreads cell damage faster. Comparisons showed unhealthy joints had only half the antioxidants in them as healthy joints.

Cancer–Numerous studies show oxidative stress affects a cell’s DNA to initiate and progress every stage of cancer: Cancer cell initiation Tumor growth Tumor spread Resistance to chemotherapy

Alzheimer’s –When inflammation develops in the brain, it generates high levels of free radicals that help create more of the amyloid plaques associated with Alzheimer’s disease. Then the free radicals generate more inflammation, which in turn creates more free radicals and consequently, more amyloid plaques. This kills neurons faster and quickens the spread of the disease.

Unfortunately, even healthy people need to worry about oxidative stress. Because it’s estimated free radicals attack your cells every 10 seconds. That’s… 600 quadrillion attacks (600 with 15 zeroes after it) a day terrorizing your body and brain!

“…we may be on the verge of a new literature on health effects of Nrf2 which may well become the most extraordinary therapeutic and most extraordinary preventive breakthrough in the history of medicine."

NRF2 physiologic action

IMPORTANCE OF CELLULAR PROTECTION

The many roles of Nrf2 The Nrf2 signaling pathway is emerging as a critical and principal factor in the protection Against cancer Neurodegeneration Inflammation Cellular detoxification Loss of Nrf2 with age is reversible and therefore an attractive target for antiaging interventions.

PULSATILE VS CONSTITUTIVE NRF2 UPREGULATION

MITOCHONDRIA AND NRF1 Mitochondria are the principal energy sources of the cell that convert nutrients into energy through cellular respiration Most estimates suggest that the majority of intracellular ROS production is derived from the mitochondria.

NRF1 NRF1 is a transcription factor which activates the expression of some key metabolic genes regulating cellular growth and nuclear genes required for respiration, heme biosynthesis, and mitochondrial DNA transcription and replication. The protein has also been associated with the regulation of neurite outgrowth. NRF-1 is a part of the adaptation of skeletal muscle to exercise training (Murakami et al. 1998; Baar et al. 2002).
