2 Introduction In this experiment we tried to look how pH can control the color of indicator. We also looked at the effect of urease from soya beans on urea. For the last experiment we were focus on how substrate's concentrations affect the enzyme's work. In procedure 1, we predicted that acid would change to red, base changed to purple and the neurtral would have no change. From procedure 2, the test tube which has both water and enzyme the color will not change, and for the one that have urea and urease the color will change. And the one that we had choose, which is substrate concentration we predicted that the one that have the most concentration can change the color easier than others.

3 Question 1. Why is it important to have three reference tubes?  To check if the indicator can work with every states, acid, base and neutral. 2. Can you predict what color of the solution will be if the urease enzyme convert urea to ammonia?  Blue or purple because ammonia is base.

4 Question 4. What are the purposes of procedure 2? To check if the enzyme is working or not. If the enzyme works probably, the tube which has both urea and enzyme must change the color to purple. 5. Why do we need to add an enzyme into a test tube without urea even though we can predict that no enzyme activity will occur? Because we need to check if enzyme is neutral. If this test tube change color then it mean that enzyme is not neutral and urea is not relate with it.

5 Question 6.How does substrate concentration affect the urease activity? How do you know? The one which has the high concentration will has high rate, take less time, to change into ammonia because from our experiment the one which has highest concentration has the most smelly. 7. Why do you think the color of the indicator slowly changes as times passes? Because it take times to make urea react with water and produce ammonia which is base.

6 Question 8.If the concentration of the enzyme was to be increased to twice the amount that was used, what do you think will happen to the color change relative to time? Explain possible reasons to support your answer. The color would change faster because the rate of changing urea to ammonia increases but it will faster until some point which is the maximum point that there are not enough urea to change to ammonia.
