2 In this experiment help us to investigate the enzyme urease by using the temperature to observed the substrate concentrations affect enzyme activity.

3 Procedure 1 -2 drops of the critic acid with indicator change their color to light pink. -2 drops of baking soda with the indicator made the color change from purple to blue. -2 drops of dH20 made the indicator change their color to become lighter.

12 Result after adding Urea and Indicator
When return into the test tube: Test tube in ice bath: Stay the same (purple) Test tube at room temperature(25°C): Does not denatured and change from purple to green Test tube in water bath (70°C): Not all denatured (We can see that active site can process). Test tube at 50°C temperature (45°C): Enzyme not denatured, change from purple to green Test tube in boiling water: Enzyme denatured, stay the same

13 Observed the color change after 5 minutes
Test tube in ice bath: Turned from purple to light blue. We figured out that enzyme working when the temperature get warmer. Test tube at room temperature(25°C): Does not denatured and change from green to dark green Test tube in water bath (70°C ): Not all denatured, turned to green (We can see that active site can process). Test tube at 50°C temperature (45°C): Enzyme not denatured, change from green to dark green Test tube in boiling water: Enzyme denatured, stay purple.

14 Conclusion -We have learn how much temperature will kill enzyme by putting 4 test tube in 4 different temperatures. -It have some like when put enzyme at room temperature the bigger was open stove first then it make the temperature go very high we need to be careful before doing experiment -The prediction that we make before was enzyme will be survive in every temperature but it doesn't when we make experiment it can't be survive at 100 degree Celsius and at 0 degree Celsius it was died but not all of them.
