Lyndon B. Johnson Space Center - Houston, TX CELLULAR BIOTECHNOLOGY OPERATIONS SUPPORT SYSTEM Biotechnology Specimen Temperature Controller (BSTC) Functional Check-Out Overview Includes Gas Supply Module (GSM)

Lyndon B. Johnson Space Center - Houston, TX CELLULAR BIOTECHNOLOGY OPERATIONS SUPPORT SYSTEM Lesson Objectives At the completion of this Systems Overview session, the Crew should be able to: Know the purpose of the Biotechnology Specimen Temperature Controller (BSTC) and Gas Supply Module (GSM) hardware Understand the set up and functional checkout of the BSTC and GSM hardware Understand the operations and crew actions required to complete the BSTC Functional Check-out.

Lyndon B. Johnson Space Center - Houston, TX CELLULAR BIOTECHNOLOGY OPERATIONS SUPPORT SYSTEM The BSTC is an incubator that provides the proper thermal and gas environment for growth and proliferation of three-dimensional cell cultures in microgravity. It is Single mid-deck locker EXPRESS Rack payload (requires EXPRESS power and data connections). It is designed to take air in through the front panel and exhaust it through vents at the side of the locker. (Front Breather) It contains a single Incubation Chamber which operates in heat only mode enclosed by two doors that maintain temperature and pressure. The BSTC contains a touch panel / LCD display for data entry and viewing system parameters. It includes a PCMCIA drive, requiring two PCMCIA cards, accessible from the front panel behind a Drive Cover, held on by thumb screws. Consumes a maximum of 100W of power at start up and 56W while incubating (power is approximately 28 W while idle). BSTC Purpose and Design

Lyndon B. Johnson Space Center - Houston, TX CELLULAR BIOTECHNOLOGY OPERATIONS SUPPORT SYSTEM GSM Purpose and Design The GSM is designed to supply mixed gas to support payloads in need of maintaining metabolic parameters and physiological balance of cell culture media. The GSM contains two independent Gas Supply Systems, 1 and 2. Each Gas Supply System has a corresponding outlet valve (one) and two Supply Valves from two cylinders containing CO 2. The hardware is packed in an ISS stowage locker with foam isolation material covering gas cylinders.

Lyndon B. Johnson Space Center - Houston, TX CELLULAR BIOTECHNOLOGY OPERATIONS SUPPORT SYSTEM BSTC Functional Check-Out OPERATIONAL FLOW & CREW TIME BSTC-ACTIVATE-GSM – crew activates GSM (0:15) BSTC-ACTIVATE – crew activates BSTC (0:30) BSTC-PARAMETERS – crew notes BSTC parameters for call down. Perform twice, near beginning and end of Ops (0:05) BSTC-DEACTIVATE – crew deactivates BSTC (0:45) PRO-BSTC-CMD – PRO commanding to prepare for activation (1:30) PRO-BSTC-CMD – PRO commanding after deactivation (1:30) BSTC-OPS – Tracks power and data during checkout (1:00).

Lyndon B. Johnson Space Center - Houston, TX CELLULAR BIOTECHNOLOGY OPERATIONS SUPPORT SYSTEM BSTC Functional Check-Out OPERATIONAL FLOW & CREW TIME 1 st Activity: 2.010 BSTC Activate GSM Crew Time: 15 min Check Outlet Valves CLOSEDOPEN Refer to Execution Notes to understand which Gas Supply System to manipulate.

Lyndon B. Johnson Space Center - Houston, TX CELLULAR BIOTECHNOLOGY OPERATIONS SUPPORT SYSTEM 2 nd Activity: 2.001 BSTC Activate Crew Time: 30 min Embedded Procedures: 2.011 BSTC Connect Gas Supply Hose 2.002 BSTC Access 2.003 BSTC Close When accessing BSTC Inner Chamber Door, note the inner door is not secured on the hinges. It is intended to release from the BSTC and has a velcro strip for quick tmpry stow. 3 rd Activity: 2.041 BSTC Parameters Crew Time: 05 min (performed twice for total of 10 min) Crew will notate BSTC Parameters at the beginning of the hour and at the end of the hour. Crew will call down data at their convenience. BSTC Functional Check-Out OPERATIONAL FLOW & CREW TIME

Lyndon B. Johnson Space Center - Houston, TX CELLULAR BIOTECHNOLOGY OPERATIONS SUPPORT SYSTEM 4 th Activity: 2.009 BSTC Deactivate Crew Time: 45 min Embedded Procedures: 2.013 BSTC Disconnect Gas Supply Hose 2.012 BSTC Deactivate GSM 2.002 BSTC Access 2.003 BSTC Close BSTC Front Panel will display a touch screen upon powering up. Delete Log Files by selecting “Settings”. Crew will stow (2) BSTC Data Cables, (2) GSM Gas Supply Hoses, (4) BSTC Program PC Cards and (1) Subnet Mask PC Card in the inner chamber of the BSTC. All components can withstand temperatures reached during Check-out; cooling of the inner chamber before stow is not necessary. BSTC Functional Check-Out OPERATIONAL FLOW & CREW TIME
