Notes 12.1 & 12.2 Early DNA Discoveries Griffith 1928 & Avery 1944: DNA (not RNA) is transforming agent. Hershey-Chase 1953: DNA (not protein) is the genetic material. Gierer & Schramm 1956/Fraenkel-Conrat & Singer 1957: RNA (not protein) is genetic material of some viruses, but no known prokaryotes or eukaryotes use RNA as their genetic material. Alfred Hershey Nobel Prize in Physiology or Medicine 1969 8

Structure of DNA James D. Watson/Francis H. Crick 1953 proposed the Double Helix Model based on two sources of information: 1.Base composition studies of Erwin Chargaff indicated double-stranded DNA consists of ~50% purines (A,G) and ~50% pyrimidines (T, C) amount of A = amount of T and amount of G = amount of C (Chargraff’s rules) %GC content varies from organism to organism Examples:%A%T%G %C%GC Homo sapiens 31.031.519.118.437.5 Zea mays 25.625.324.524.649.1 Drosophila 27.327.622.522.545.0 Aythya americana 25.825.824.224.248.4 9

Notes 12.3 DNA polymerase is an enzyme that joins individual nucleotides to produce a new strand of DNA. Replication in most prokaryotic cells starts from a single point and proceeds in two directions until the entire chromosome is copied. In eukaryotic cells, replication may begin at dozens or even hundreds of places on the DNA molecule, proceeding in both directions until each chromosome is completely copied.
