1 Avena genetic resources for quality in human consumption (AVEQ)
Partner 12 IHAR -PIB Laboratory of Quality Evaluation of Plant Materials Institute of Plant Breeding and Acclimatization - National Research Institute, Radzików, PL , Poland AVEQ Final Meeting – Bucharest October 19-21, 2010

2 WP 7. Task 1, Sub-task 4 . Analyses for starch and dietary fibre
The objective: Characterise carbohydrates - starch and dietary fibre in genetic resources of Avena. AVEQ Final Meeting – Bucharest October 19-21, 2010

3 WP 7. Task 1, Sub-task 4 . Analysis of starch - selected method
-amylase/amyloglucosidase method: ICC Standard no. 128/1and168 or AACC 76-13 or AOAC AVEQ Final Meeting – Bucharest October 19-21, 2010

4 WP 7. Task 1, Sub-task 4 . Analysis of dietary fibre - selected method
Enzymatic-Chemical Uppsala Method (AOAC or AACC 32-25) Gas chromatographic-colorimetric-gravimetric procedure TDF = neutral sugar residues + uronic acids + Klason lignin Principle 1. Measurement of NSP as neutral sugar residues by GLC (Englyst and Cummings procedure, 1988) starch removal by specific enzymes precipitation of soluble polymers by ethanol hydrolysis of polysaccharides to monosaccharides with H2SO4 reduction and derivatisation 2. Measurement of uronic acid by colorimetry (Scott method, 1979) 3. Measurement of Klason lignin by gravimetric method as the sulphuric acid resistant residue (Theander et al.,1995)

5 Dietary Fibre Constituents
I. Non-starch polysaccharides & oligosaccharides Cellulose Non-cellulosic polysaccharides Polyfructoses inulin oligofructans Galactooligosugars Hemicelluloses arabinoxylans arabinogalactans β-glucans Pectins Gums Mucilages IV. Substances associated with NSP & lignin complex in plants II. Analogous Carbohydrates Waxes Phytates Cutin Suberin Saponins Tannins Resistant starch Synthesized carbohydr. Indigestible dextrins III.
