3 MERITS OF CONFOCAL MICROSCOPY a)the ability to serially produce thin (0.5 to 1.5 micrometer) optical sections through fluorescent specimens that have a thickness ranging up to 50 micrometers or more. b)Image information is restricted to a well-defined plane, rather than being complicated by signals arising from remote locations in the specimen. c)Contrast and definition are dramatically improved over widefield techniques due to the reduction in background fluorescence and improved signal-to-noise. d)eliminates artifacts that occur during physical sectioning and fluorescent staining of tissue specimens for traditional forms of microscopy.

6 APPLICATIONS: a)widely-used in biological science disciplines, from cell biology and genetics to microbiology and developmental biology. b)evaluation of various eye diseases, and is particularly useful for imaging, qualitative analysis, and quantification of endothelial cells of the cornea c)endoscopic procedures: In the pharmaceutical industry, it was recommended to follow the manufacturing process of thin film pharmaceutical forms, to control the quality and uniformity of the drug distribution. d)data retrieval mechanism in some 3D optical data storage systems and has helped determine the age of the Magdalen papyrus.
