2 Figure 1 Regulation of hepcidin expression in hepatocytes. The expression of hepcidin is positively regulated by diferric transferrin (Tf(Fe2)), inflammation and by receptors for bone morphogenetic proteins (BMPs). Hepcidin is negatively regulated by anemia and hypoxia. The activation of cell-surface BMP receptors (BMPsR) by BMPs leads to the generation of phosphorylated receptor SMADs (SMADs-P). Membrane-bound hemojuvelin (HJV) functions as a co-receptor for BMPs, increasing the efficiency of the interaction of BMP with its receptor. Tf(Fe2) induces hepcidin via transferrin receptor-2 and HFE through an independent signal or by affecting signal transduction by the BMPsR.
