1 Mir-128 reduction in breast cancer tissues correlates chemotherapy resistance and poor patient survival. Mir-128 reduction in breast cancer tissues correlates chemotherapy resistance and poor patient survival. MiR-128 expression was reduced in chemoresistant breast cancer tissues but upregulated in chemosensitive tissues, according to in situ hybridization (A) and real-time RT-PCR with a miR-128–specific primer amplifying both miR-128a and miR-128b (B, **, P < 0.001). A, representative pictures of immunohistochemical staining for Bmi-1 and ABCC5 in breast cancer tissues. Bmi-1 and ABCC5 expression was higher in chemoresistant than in chemosensitive breast tumor tissues. C, representative pictures of immunohistochemical staining for Ki67 (×200) and TUNEL staining (×200) in paired specimens of breast cancer obtained by core-needle biopsy prior to neoadjuvant chemotherapy and at surgery following chemotherapy. Breast tumors with low miR-128 expression showed stable Ki67 index and TUNEL index following chemotherapy, whereas those with high miR-128 showed a dramatically reduced Ki67 index and elevated TUNEL index after chemotherapy. D, Kaplan–Meier survival curves for breast cancer patients plotted against miR-128 expression show a statistical difference in survival between patients with high- and low-miR-128 expression (P < 0.05).
