Radiosensitization by combined Wee1 and PARP inhibition. AsPC-1 (A) and MiaPaCa-2 (B) pancreatic cancer cell lines were treated with AZD1775 (100 nmol/L), olaparib (1 μmol/L), and radiation (RT; 0–6 Gy) according to the schedule illustrated (D). At the end of the treatment, cells were processed for clonogenic survival. Data are from a single representative experiment (A and B) or are the mean radiation enhancement ratio (RER) ± SE for n = 3 independent experiments (C). Cytotoxicity in the absence of radiation treatment was calculated by normalizing the plating efficiencies of drug-treated cells to non–drug-treated cells where plating efficiencies for non–drug-treated AsPC-1 and MiaPaCa-2 cells were 0.2 and 0.3, respectively (C).
