Cell viability assay and expression of COX protein. A, in the cell viability assay, colon cancer cell lines were grown in a monolayer for 5 days with 0.01% DMSO (control) or with 50 μm or 100 μm NS398. The cells were fixed with TCA and stained with SRB. Absorbance was read on an automated spectrophotometric plate reader at 490 nm and percentage of growth inhibition (GI) was calculated. The experiments were in triplicate and repeated once. B, results of quantitative analysis of COX-2 protein from Western blotting using the IS-1000 digital-imaging system (Alpha Innotech Corp., San Leandro, CA).
