You should be able to … 1. describe gene expression by explaining the following: a. transcription b. translation 2. given a DNA sense strand give the following: mRNA strand and polypeptide (amino acid sequence). 3. Describe how the lac operon works to break down lactose. Be sure to cover both repression and activation. Include: ligand (inducer), regulator and structural genes, promoter, repressor, operator, RNA polymerase. 4. explain why Escherichia coli is a great organism to use for the transformation process. 5. explain how Griffith discovered the transformation process using mice and a virulent strain of bacteria. 6. compare the lac operon to the arabinose operon by explaining similarities and differences. 7. describe how the arabinose operon was engineered to transform our Escherichia coli cells. 8. describe how an Escherichia coli cell is transformed. (i.e. What did you do in the lab to cause the cell to uptake the pGLO plasmid?) a. explain why the CaCl2 was used. b. explain why the heat shock was used. 9. define the following “key terms” as they pertained to the objective of the transformation lab: plasmid, colony, ampicilin (antibiotic), arabinose, GFP protein, arabinose operon, and transcription. 10. why was the ampicilin used in this lab on both the control and experimental groups?
